Amy Schumer was born in Manhattan and spent her formative years on Long Island, New York. She attended South Side High School, where she earned the titles of "Class Clown" and "Teacher's Worst Nightmare" upon her graduation in 1999. In 2003, Schumer completed her theater degree at Towson University. Following her college years, she relocated to New York City, where she honed her craft for two years at the William Esper Studio while balancing jobs as a bartender and waitress. She is also connected to U.S. Senator Chuck Schumer, as he is her uncle.

In her early career, Schumer portrayed a young woman coping with a breast cancer diagnosis in the off-Broadway dark comedy, Keeping Abreast. She ventured into stand-up comedy on June 1, 2004, with her debut performance at Gotham Comedy Club. Her big break came when she recorded a special for Comedy Central, which she later viewed as a pivotal moment in her career.

Though she initially did not succeed in an audition for an earlier season, Schumer made it to the finals of the fifth season of NBC's reality talent show Last Comic Standing, finishing in fourth place. Recalling the experience in April 2011, she remarked that the competition was enjoyable and low-pressure, as she had only been performing stand-up for about two years. Her authenticity during the show contributed to her success.
