Patinkin's theatrical journey began in 1975 with a notable debut opposite Meryl Streep in the revival of Trelawny of the "Wells" at The Public Theatre's Shakespeare Festival. He gained acclaim for his portrayal of Che in the original Broadway production of Andrew Lloyd Webber's Evita (1979), winning a Tony Award for Best Featured Actor in a Musical. His role as Georges Seurat/George in Sondheim's Sunday in the Park with George (1984) earned him a Tony nomination for Best Actor in a Musical, while he also played Lord Archibald Craven in the original cast of The Secret Garden (1991). Patinkin starred as Burrs in The Wild Party (2000) and returned to Broadway as Marvin in Falsettos (1993).

In television, he is recognized for his roles as Dr. Jeffrey Geiger in Chicago Hope (1994–2000), SSA Jason Gideon in Criminal Minds (2005–2007), and Saul Berenson in Homeland (2011–2020), winning an Emmy for Chicago Hope.
