Marlene Willoughby adopted her professional surname after being described as "willowy." Inspired by her older sister, Jacqueline Carol, she ventured into the entertainment industry. In 1969, she made her mark in the provocative Off-Off-Broadway production Che!, directed by Lennox Raphael. This was just the beginning of her theatrical journey, which included acclaimed performances in the Obie Award-winning play Dracula Sabbat, as well as Fuck Mother and Keepers of the Hippo Horn.

Willoughby transitioned to film with roles in notable mainstream projects like No Place to Hide (1970), Up the Sandbox (1972), I, the Jury (1982), and Trading Places (1983). She also participated in the softcore genre, featuring in titles such as Voices of Desire (1972) and While the Cat's Away... (1972). In 1975, she entered the adult film industry, gaining recognition for her work in classics like The Opening of Misty Beethoven (1976), The Farmer's Daughters (1976), Outlaw Ladies (1981), and Foxtrot (1982).

Beyond acting, Willoughby contributed to various adult magazines, notably Penthouse, and penned columns for High Society and Velvet. She organized film release events and toured as a burlesque performer.
